  • Mercury-induced dark-state instability and photobleaching alterations of the visual G-protein coupled receptor rhodopsin

    Mercuric compounds were previously shown to affect the visual phototransduction cascade, and this could result in vision impairment. We have analyzed the effect of mercuric chloride on the structure and stability of the dim light vision photoreceptor rhodopsin. For this purpose, we have used both native rhodopsin imrnunopurified from bovine retinas and a recombinant mutant rhodopsin carrying several Cys to Ser substitutions in order to investigate the potential binding site of mercury on the receptor. Our results show that mercuric chloride dramatically reduces the stability of dark-state rhodopsin and alters the molecular features of the photoactived conformation obtained upon illumination by eliciting the formation of an altered photointermediate. The thermal bleaching kinetics of native and mutant rhodopsin is markedly accelerated by mercury in a concentration-dependent manner, and its chromophore regeneration ability is severely reduced without significantly affecting its G-protein activation capacity. Furthermore, fluorescence spectroscopic measurements on the retinal release process, ensuing illumination, suggest that mercury impairs complete retinal release from the receptor binding pocket. Our results provide further support for the capacity of mercury as a hazardous metal ion with reported deleterious effect on vision and provide a molecular explanation for such an effect at the rhodopsin photoreceptor level. We suggest that mercury could alter vision by acting in a specific manner on the molecular components of the retinoid cycle, particularly by modifying the ability of the visual photoreceptor protein rhodopsin to be regenerated and to be normally photoactivated by light.

    Rhodopsin and cone opsins, the visual pigments of the vertebrate retina are a distinct group of G-protein-coupled receptors which are covalently bound to their ligand, the chromophoric 11-cis-retinal in the resting inactive state. The retinal binding site of these receptors is very dynamic allowing their conformational stabilization and functional activation. Though various retinal analogs were reported to behave as chromophores for retinal receptors, their specific physical properties and activation ability varies among them. We have carried out a study of the dynamics of the retinal binding site of rhodopsin and cone opsins after photo-activation which had not been previously analyzed. In order to do that, the photoreceptor genes inserted into the pMT4 vector were transiently transfected in COS-1 cells. Opsins regenerated with 11-cis-retinal were immunopurified by using rho-1D4 antibody. Fluorescence spectroscopy was employed to monitor conformational changes upon ligand binding to the receptor with retinal analogs 11-cis-retinal and 9-cis-retinal and their functional behavior was measured using a G-protein activation assay. We found that, after photo-activation, both retinals can access the binding pocket of red cone opsin but only 9-cis-retinal can occupy the retinal binding site of rhodopsin. The binding kinetics of 9-cis-retinal to photo-activated rhodopsin showed a faster entry into the retinal binding site followed by a binding process with t1/2 of ~1 min. Furthermore, the G-protein activation profile of post-bleached red cone and rhodopsin reveal differences which may account for the different rod and cone sensitivities. In summary, the present study suggests that the rhodopsin retinal binding site is less conformationally flexible than the red cone binding pocket which may be more open to permit the entry of both retinal analogs.

    El treball que es presenta forma part del Projecte Elaboració de material per a l'Avaluació d'assignatures de Química, de la convocatòria d'ajuts per a projectes de Millora de la docència 2009-2010 de la Universitat Politècnica de Catalunya. Els autors pertanyen als Grups GReDiQ i GRAPA integrats al projecte RIMA de l’Institut de Ciències de l’Educació de la UPC. S’ha elaborat un banc de preguntes i problemes, accessible a tot el professorat de la UPC que imparteix assignatures de Química i que podrà ser utilitzat per posar en pràctica l’avaluació continuada de l’aprenentatge: autoavaluació formativa i avaluació sumativa. Hi ha qüestionaris Moodle referents a la utilització del material multimèdia generat en un projecte anterior (“Qüestionaris de laboratori”) i d’altres de problemes (“Qüestionaris de problemes”). Es pretén valorar la repercussió del material multimèdia en l’aprenentatge i l’assoliment dels objectius formatius. Els qüestionaris de laboratori s’han utilitzat com part del pre-laboratori de l’alumnat de Experimentació en Química II (ETSEIB), Experimentació en Química (ETSEIB), Química (EET) i Anàlisi Química (EET). Els qüestionaris de problemes s’han utilitzat en les assignatures de Química i Anàlisi Química de l’EET i s’estan implementant aquest quadrimestre a la FOOT (a Química per a ciències de la visió) tant a la modalitat presencial com semipresencial. L’avaluació dels primers resultats es va realitzar durant el primer quadrimestre del curs 2011-12 mitjançant una enquesta de satisfacció que es va passar a l’EET. L’alumnat de l’EET utilitza majoritàriament els qüestionaris i els troben útils per tal de valorar el seu progrés a l’assignatura.

    En aquesta comunicació es presenta un projecte elaborat arran de l’interès mostrat per un grup de professorat en disposar de material audiovisual propi i comú per a assignatures experimentals de química de diferents titulacions impartides per diferents centres de la Universitat Politècnica de Catalunya (UPC). Els autors d’aquest treball pertanyen al Grup de Recursos per a la Didàctica de la Química (GReDiQ), integrat en el projecte RIMA (http://www.upc.edu/rima) de l’Institut de Ciències de l’Educació de la UPC. L’elaboració de material audiovisual digital constitueix un dels objectius principals plantejats pel GReDiQ i pretén generar tant un material de suport per a les activitats pre-laboratorio, com un element motivador i que faciliti l’aprenentatge de l’alumnat en les sessions de laboratori de les matèries de Química i Enginyeria química, que apareixen com assignatures de formació bàsica en gairebé tots els plans d’estudi dels nous Graus d’Enginyeria que s’ estan implantant a la UPC. La utilització del material audiovisual digital possibilita que l’alumnat prengui consciència de les activitats de laboratori, així com dels hàbits bàsics de manipulació i seguretat en el laboratori, del fonament d’operacions bàsiques o instrumentació a utilitzar.

    Se han obtenido y caracterizado una serie de poli(a-n-alquil g-glutamato)s (PAAG-n) con n³12 y de poli(a-n-alquil b,L-aspartato)s (PAALA-n), mediante transesterificación con n-alcoholes y se ha estudiado la influencia que ejerce el grupo lateral sobre la estructura y propiedades de estos polímeros. Los polímeros se han caracterizado mediante FTIR, RMN de 1H y 13C y análisis elemental. La composición enantiomérica se ha determinado midiendo rotaciones específicas de los polímeros hidrolizados y/o mediante HPLC. Los pesos moleculares se han evaluado por viscosimetría y/o SEC/LS. La caracterización térmica se ha llevado a cabo mediante DSC, TGA y DMTA. El análisis estructural se ha realizado mediante las técnicas de DIR, 13C-CP/MAS-RMN, difracción de rayos X y de electrones. Los PAAG-n se han obtenido a partir del ácido poli(g-glutámico) (PGGA) de biosíntesis, de dos composiciones enantioméricas diferentes, uno enriquecido en el isómero D y otro de composición cercana a la racémica. El procedimiento general está basado en dos reacciones sucesivas (esterificación y transesterificación) y ha resultado especialmente útil para la preparación de ésteres de cadena larga difíciles de obtener por otros métodos. Las conversiones han sido completas no observándose racemización significativa. Los PAAG-n con n³14 forman una estructura bifásica supramolecular en láminas. Las cadenas principales adoptan una conformación helicoidal tipo hélice a y se alinean en láminas con las cadenas laterales interdigitadas formando una microfase entre las láminas. La distancia interlaminar es proporcional a n. Para estos polímeros se han encontrado las transiciones de fase A®B y B®C, a temperaturas T1 y T2. La transición A®B es reversible y tiene lugar a temperaturas entre 20 y 80 ºC. La transición B®C se ha observado para los PAAG-n con n³18 a temperaturas superiores a los 100 ºC. En la fase A las cadenas laterales están cristalizadas en una red hexagonal y dispuestas perpendicularmente a la cadena principal. En la fase B las cadenas laterales están desordenadas pero más o menos estiradas. La fase C presenta las cadenas laterales totalmente desordenadas. No se han observado efectos termocrómicos al calentar las muestras desde temperatura ambiente hasta temperaturas superiores a T2 cuando se observan bajo luz polarizada.Los PAALA-n y el PIBLA (poli(a-isobutil b,L-aspartato)) se han obtenido por transesterificación del PABLA (poli(a-bencil b,L-aspartato)). Resultó difícil conseguir conversiones completas debido a la existencia de reacciones de imidación. Los PAALA-n obtenidos mediante transesterificación mostraron temperaturas de descomposición similares a las de los PAALA-n obtenidos mediante polimerización. Las transiciones a y b (DMTA) se observan en los PAALA-n de cadenas cortas e intermedias a temperaturas más elevadas que las descritas para los PAAG-n con el mismo valor de n. La Ta disminuye a medida que aumenta n. La Tb aumentó ligeramente con n para los PAALA-n con n£ 8. La relajación g se observa para los PAALA-n con n³6.
